At the Louvre Museum in Paris, curator Jacques Saunière locks himself in the Grand Gallery, pursued by an intruder — an albino monk named Silas, who demands the location of a stolen object. When Saunière lies, Silas shoots him in the stomach and leaves. Realizing his fellow secret-keepers have already been killed, Saunière knows he must pass on the knowledge entrusted to him before he dies.

At the Hôtel Ritz Paris, Harvard symbology professor Robert Langdon is woken and taken to the Louvre to help decipher the bizarre clues left behind by the dying curator. DCPJ cryptographer Sophie Neveu arrives with news that Saunière's scrawled numbers form a scrambled Fibonacci sequence — and quietly informs Langdon that he's the lead suspect, since Saunière also wrote his name on the gallery floor. Sophie, revealed to be Saunière's estranged granddaughter, is convinced he's left her a coded message about her deceased family, and she helps Langdon escape the Louvre, believing he can help her follow the trail.

As Langdon and Sophie sort through the riddles, they discover a trail of clues hidden in the works of Leonardo da Vinci — clues visible for all to see yet ingeniously disguised by the painter. Their search leads them to Sir Leigh Teabing, a Grail historian, and reveals that the late curator was a member of the Priory of Sion — a secret society whose past members allegedly included Isaac Newton, Victor Hugo, and da Vinci himself, guarding a historical secret of explosive significance. The novel proposes that Jesus and Mary Magdalene were secretly married and had a child together, continuing a bloodline the Priory has protected for two thousand years.

All the while, the pair are pursued by the police and by Silas, who is affiliated with Opus Dei, a conservative Catholic organization also entangled in the conspiracy. Silas, acting on the orders of a mysterious figure known only as the Teacher, continues to hunt down the remaining secret-keepers to retrieve a keystone that holds the key to finding the Grail. Through a series of intricate puzzles, codes, and anagrams, Langdon and Sophie unravel the mystery step by step until, in the climactic final act, they uncover the location of the Holy Grail and confront the true identity of the person behind the murders and the conspiracy — a shocking twist that ties every thread of the story together.
